The Trigun anime takes place across a number of different locations on No Man's Land. This page provides an overview, with links where necessary to provide greater detail.
Major Cities
There are seven major cities in Trigun lore, though only four are mentioned in the anime:

The city that earned Vash his gigantic bounty.
Augusta
The city that Vash is goaded by Legato into annihilating.
May City
Wolfwood gets off the bus here. They also host the 3rd annual Quick Draw tournament.
December City
Where Bernardelli HQ is located, and where Meryl is from.

Lob's Diner is 10 iles from here.
Dankin Town
Gets obliterated at the end of Episode 1.
Lewiston Town
Milly and Meryl stay here briefly, while figuring out how to continue looking for Vash.
Orleans
Where "Grim Reaper" Bostalk and his daughter Stefany live.
LR Town
Vash fights with Midvalley here.
Promontory
The city in "Little Arcadia".
Little Arcadia
An oasis in the middle of the desert, likely sustained by a geoplant.
Denislad
A small town adjacent to a geyser field, through which caravans pass.

Fondrique
A caravan town involved in human trafficking.

Where Vash faces off against the Nebraskas.

Where the Polo/Fris brutality happens.
Keybos
A walled town trying to protect itself from thugs and sandworms.
Karsted City
Where Lina and her grandmother live.

A town whose residents all mysteriously vanished one day.
Tonim Town
Where Wolfwood is confronted by Chapel.
Demitrihi
Where Knives is. Possibly also where Milly and Meryl take Vash to recuperate after the events of episode 24.
